Overview

The CT-26 WT vaccine is a preclinical cancer vaccine derived from the CT-26 murine colon carcinoma cell line. In the context of the described research, the vaccine consists of CT-26 cells that have been genetically modified to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF) and subsequently irradiated to prevent further proliferation while maintaining metabolic activity and antigenicity. This approach utilizes the whole tumor cell to provide a diverse array of tumor-associated antigens to the host immune system. The secreted GM-CSF serves as a potent local adjuvant, enhancing the recruitment, activation, and maturation of dendritic cells at the injection site. These dendritic cells then process the tumor antigens and present them to T cells, inducing a systemic antitumor immune response. This vaccine has been studied for its ability to provide protective immunity and therapeutic efficacy against liver metastases in syngeneic BALB/c mouse models.
