Overview

CT-47609 (also known as CT0109) is a small molecule Sigma-2 receptor (S2R) antagonist developed by Cognition Therapeutics for the treatment of Alzheimer's disease. It acts as a high-affinity ligand for the Sigma-2 receptor complex, which serves as a binding site for toxic amyloid-beta (Aβ) oligomers. By competitively inhibiting the interaction between Aβ oligomers and the receptor complex at the synapse, CT-47609 prevents oligomer-induced synaptic dysfunction and facilitates the displacement of pre-bound oligomers. This mechanism is intended to halt the progression of cognitive decline by protecting neuronal signaling and synaptic density. While CT1812 has advanced as the lead clinical candidate for Cognition Therapeutics, CT-47609 remains a key compound in the characterization of Sigma-2 modulation as a therapeutic strategy for neurodegenerative disorders.
