Overview

**CT-L01** is a Celltrion fixed-dose oral tablet containing the dipeptidyl peptidase-4 inhibitor **alogliptin** and **extended-release metformin**. It is being developed as a combination treatment for type 2 diabetes mellitus. Alogliptin prolongs endogenous incretin activity through selective, reversible inhibition of dipeptidyl peptidase-4, thereby increasing glucose-dependent insulin secretion and reducing glucagon secretion. Metformin primarily reduces hepatic glucose production and improves insulin sensitivity; its molecular actions are associated with inhibition of mitochondrial respiratory-chain complex I and downstream activation of AMP-activated protein kinase. Registered Phase 1 studies evaluated bioequivalence of CT-L01 against co-administered alogliptin plus metformin extended release and evaluated food effects in healthy volunteers.
