CT-P72 + ABP-102 is a **tetravalent bispecific antibody** designed as a next-generation immuno-oncology therapy targeting HER2 and CD3. Engineered using dual-affinity tuning and the “TetraBi” format, the molecule features bivalent binding domains for HER2 and a functionally monovalent affinity-tuned domain for CD3. This design allows for potent, selective binding to HER2-overexpressing (HER2+) tumor cells, redirection and activation of cytotoxic T cells through CD3 engagement, and reduced activity on HER2-low and HER2-negative normal tissues, greatly minimizing on-target, off-tumor toxicity and cytokine-related side effects. Preclinical data demonstrate robust T-cell activation, strong anti-tumor cytotoxicity, and a favorable safety profile in cynomolgus monkeys, including efficacy against breast, gastric, colorectal, and other HER2-positive tumors, including those resistant to existing antibody-drug conjugates (ADCs) such as Enhertu. CT-P72 + ABP-102 is co-developed by Abpro and Celltrion, with clinical trials planned for HER2+ breast, gastric, pancreatic, colorectal, and other cancers[1][2][3][4][5][6][7][8][9][11].
