Overview

CT0181 is an autologous T-cell therapy developed by CARsgen Therapeutics for the treatment of advanced hepatocellular carcinoma (HCC). It is engineered to express a fusion protein that combines a Glypican-3 (GPC3)-targeted antibody and a T-cell receptor, with co-expression of the cytokine IL-7. The primary mechanism involves targeting Glypican-3 (GPC3), which is highly expressed in liver cancer cells. Preclinical studies have shown that CT0181 offers superior antitumor efficacy, enhanced T-cell persistence, and improved immunological memory compared to conventional GPC3 CAR-T cells, while also exhibiting low cytokine release. A Phase I clinical trial has been initiated in China to evaluate its safety and efficacy in patients with advanced HCC[2][6][8].
