CT0991 is an autologous chimeric antigen receptor (CAR) T-cell therapy being developed by CARsgen Therapeutics for the treatment of hematologic malignancies. It specifically targets C-type lectin-like molecule-1 (CLL-1, also known as CLEC12A), an antigen that is highly expressed on the surface of acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs). CLL-1 is generally absent on normal hematopoietic stem cells (HSCs), providing a therapeutic window to target leukemia cells while sparing the bone marrow's regenerative capacity. The therapy involves engineering a patient's own T cells to express a CAR that recognizes CLL-1, which, upon binding, triggers T-cell activation and the targeted destruction of AML cells. CT0991 is currently under clinical investigation in patients with relapsed or refractory AML.
