Overview

CT103D is an investigational, donor-derived (allogeneic) dual-targeting chimeric antigen receptor (CAR) T-cell therapy that simultaneously targets the B-cell maturation antigen (BCMA) and CD19. Developed by IASO Biotherapeutics as an evolution of the BCMA-targeted equecabtagene autoleucel (CT103A) platform, CT103D is designed specifically for patients with relapsed or refractory multiple myeloma (RRMM) and plasma cell leukemia (PCL), particularly those who have previously undergone allogeneic hematopoietic stem cell transplantation (allo-HSCT) or whose own T-cells are unsuitable for autologous CAR-T production due to exhaustion or prior treatments. The CAR construct features fully human single-chain variable fragments (scFv) directed against both BCMA and CD19. By targeting two distinct antigens, the therapy aims to mitigate the risk of antigen escape—a common mechanism of resistance in single-target CAR-T therapies—and improve the durability of response in heavily pretreated patients. CT103D is currently undergoing Phase 1 clinical evaluation.
