Overview

cT84.66 is a chimeric monoclonal antibody of the IgG1 isotype with high specificity and affinity for carcinoembryonic antigen (CEA), a tumor-associated antigen overexpressed in several cancers including colorectal and breast cancer[4][3]. Developed by City of Hope National Medical Center, cT84.66 was designed to target CEA-expressing tumors for both imaging (radioimmunoscintigraphy) and therapy (radioimmunotherapy)[4][2]. The antibody has been radiolabeled with isotopes such as Yttrium-90 (^90Y) and Indium-111 (^111In) for these purposes[2][5]. Its mechanism involves binding to CEA on tumor cells, enabling targeted delivery of radiation or imaging agents[2][6]. Clinical trials demonstrated that cT84.66 could deliver therapeutic radiation doses to tumors but was associated with dose-limiting hematologic toxicity and immunogenicity (human anti-chimeric antibody response)[2][5]. Development has been discontinued after reaching Phase 1 clinical trials[1].
