CTA control refers to the use of the Cholera Toxin A-subunit (CTA) as a mucosal adjuvant, serving as the active control in clinical trials for the IMX101 vaccine developed by ImevaX. IMX101 is a vaccine candidate designed to prevent and treat Helicobacter pylori infection. In the context of the Phase 1a/b study (NCT02918318), CTA is administered via sublingual and intradermal routes to evaluate the safety, tolerability, and immunogenicity of the adjuvant component independently of the specific H. pylori antigens. CTA functions as a potent mucosal adjuvant by ADP-ribosylating the Gs alpha subunit of heterotrimeric G proteins, which leads to increased intracellular cAMP levels. This signaling cascade modulates the maturation and function of antigen-presenting cells, such as dendritic cells, thereby enhancing the immune response to co-administered antigens.
