CTAG1B-targeted TCR-T cells are an adoptive cellular immunotherapy consisting of primary human T cells engineered to express a high-affinity T-cell receptor (TCR) specific for the cancer/testis antigen 1B (CTAG1B). The specific TCR used in this therapy targets the CTAG1B (157-165) peptide presented by the HLA-A*02:01 MHC class I molecule. These TCRs were isolated from a high-throughput phage-displayed TCR library derived from healthy donor CD8+ T cells. CTAG1B, also known as NY-ESO-1, is an immunogenic cancer/testis antigen with expression typically restricted to germ cells but frequently overexpressed in various malignancies, including esophageal cancer. In preclinical studies, these TCR-T cells demonstrated potent antigen-dependent cytotoxicity (up to 80% lysis) and robust secretion of pro-inflammatory cytokines such as IFN-γ and TNF-α against CTAG1B-positive tumor cells. In vivo xenograft models have shown significant tumor regression and increased infiltration of effector TCR-T cells following administration.
