Overview

CTB-Tim3 scFv-expressing Lactococcus lactis is an experimental live biotherapeutic product consisting of the food-grade bacterium *Lactococcus lactis* genetically engineered to express and secrete a fusion protein. This protein comprises a single-chain variable fragment (scFv) targeting T-cell immunoglobulin-3 (Tim-3) fused to the cholera toxin B subunit (CTB), which acts as a mucosal adjuvant and carrier. Tim-3 is an inhibitory immune checkpoint molecule that, when blocked, can revitalize exhausted T-cells and enhance anti-tumor immunity. Developed as a cost-effective alternative to traditional monoclonal antibodies, this microbial delivery system has demonstrated the ability to promote lymphocyte activation and inhibit tumor growth, proliferation, and angiogenesis in preclinical models of renal cell carcinoma.
