**cTfRMAb-GDNF** is an investigational brain-penetrating recombinant fusion protein comprising human glial cell line-derived neurotrophic factor fused to the heavy chain of a chimeric monoclonal antibody against the mouse transferrin receptor. The anti-transferrin-receptor antibody domain acts as a blood-brain-barrier molecular Trojan horse, enabling receptor-mediated transcytosis after intravenous administration, while the GDNF domain activates GDNF trophic signaling in nigrostriatal dopaminergic pathways. In mouse 6-hydroxydopamine models of Parkinson's disease, systemic dosing improved behavioral and striatal tyrosine-hydroxylase measures; the construct was also evaluated in chronic mouse dosing studies.
