Overview

CTGF siRNA refers to a class of small interfering RNA (siRNA) therapeutics designed to silence the expression of connective tissue growth factor (CTGF, also known as CCN2). CTGF is a matricellular protein that plays a central role in the signaling pathways leading to fibrosis, including the induction of extracellular matrix (ECM) components like collagen. By utilizing the RNA interference (RNAi) pathway, these drugs trigger the sequence-specific degradation of CTGF mRNA, thereby reducing protein levels and inhibiting fibrotic progression. Clinical applications include the treatment of hypertrophic scars, keloids, and various organ-specific fibroses (e.g., pulmonary or liver fibrosis). Notable examples include LEM-S401, which utilizes a nanoparticle delivery system, and RXI-109, a self-delivering siRNA.
