CTH-401 is an investigational allogeneic ("off-the-shelf"), induced pluripotent stem cell (iPSC)-derived chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed by Cartherics. It is designed to target solid tumors, with a primary focus on ovarian cancer. The therapy uses NK cells engineered from iPSCs to express a proprietary second-generation CAR that specifically targets tumor-associated glycoprotein-72 (TAG-72), a marker widely expressed in various solid tumors including ovarian, gastric, colorectal, prostate, and pancreatic cancers. Additionally, two genes associated with immunosuppression are deleted to enhance the survival and efficacy of the product. The final product is derived from a clonal iPSC master cell bank and aims for precise cytotoxic action against tumor cells while resisting immunosuppressive mechanisms within the tumor microenvironment. Clinical trials for ovarian cancer are planned to begin in 2025[1][2][3][4][5][7].
