CTH120 and CTH122 are two distinct first-in-class small molecule neuroplasticity modulators developed by Connecta Therapeutics. CTH120, the lead asset, is a neuroplasticity modulator that targets the tropomyosin receptor kinase B (TrkB) pathway, a critical mediator of brain development and synaptic function. It has completed Phase I clinical trials and is progressing into Phase IIa for the treatment of Fragile X syndrome (FXS). CTH120 has also demonstrated preclinical efficacy in models of Rett syndrome, Down syndrome, and autism spectrum disorders, and holds EMA orphan drug designation for FXS. CTH122 is a newer, recently patented small molecule from the same neuroplasticity modulation platform, currently in preclinical development for FXS and Rett syndrome. Although sometimes grouped together in pipeline charts for shared indications like Rett syndrome, they are separate therapeutic candidates rather than a fixed-dose combination.
