Overview

CTH522 + CAF01 is a combination vaccine candidate designed to prevent infection by Chlamydia trachomatis, the bacterium responsible for both ocular trachoma and urogenital chlamydia. The vaccine consists of the recombinant protein antigen CTH522, which is an engineered version of the major outer membrane protein (MOMP) from Chlamydia trachomatis, formulated with the cationic liposomal adjuvant CAF01. This formulation aims to induce strong humoral and cellular immune responses, including neutralizing antibodies and multifunctional Th1/Th17 T cell responses. Preclinical studies have shown that vaccination with CTH522/CAF01 provides significant protection against reinfection in animal models by reducing bacterial load and enhancing both systemic and mucosal immunity. Clinical trials have demonstrated that this combination is safe, well-tolerated, and highly immunogenic in humans, outperforming alternative adjuvants such as aluminium hydroxide in terms of antibody production and T cell response[1][3][5][6][7][9].
