CTIM-76 is a bispecific antibody that targets both Claudin 6 (CLDN6) and CD3. It is designed as a T cell engager, incorporating a highly selective CLDN6 binding arm and a CD3 binding single-chain Fv domain in an IgG format with a silenced Fc region to minimize aberrant T-cell activation and enhance safety. The drug aims to direct cytotoxic T cells specifically against CLDN6-positive cancer cells, sparing normal tissues due to the restricted expression of CLDN6 in healthy tissue. Preclinical studies have shown potent, selective lysis of CLDN6+ tumor cells, robust anti-tumor activity in xenograft models, low immunogenicity risk, and scalable manufacturing potential. CTIM-76 is being developed primarily for the treatment of advanced or metastatic gynecologic (ovarian and endometrial) and testicular cancers that express CLDN6[1][5][7].
