Overview

CTL-05 is an experimental adoptive cell therapy consisting of autologous cytolytic T lymphocytes (CTLs) sensitized and expanded ex vivo. The manufacturing process is unique in its use of Drosophila (fruit fly) cell lines as artificial antigen-presenting cells (aAPCs). These insect cells are stably transfected with human major histocompatibility complex (MHC) molecules, specifically HLA-A2, along with necessary costimulatory and adhesion molecules (such as B7-1 and ICAM-1). When pulsed with specific melanoma-associated peptides, these Drosophila cells potently stimulate and expand tumor-reactive CD8+ peripheral blood lymphocytes (PBLs) derived from the patient. Once re-infused following a lymphodepleting chemotherapy regimen, these sensitized T cells are intended to recognize and destroy melanoma cells. The therapy was developed primarily for the treatment of HLA-A*0201-positive patients with metastatic cutaneous melanoma that has failed prior interleukin-2 (IL-2) therapy.
