CTLA-4 Vax is an experimental DNA-based vaccine designed to induce the endogenous production of antibodies against cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). By stimulating the host's immune system to generate its own checkpoint inhibitors, it aims to provide a cost-effective alternative to monoclonal antibody-based immune checkpoint blockade (ICB). In preclinical studies, particularly in murine melanoma models, CTLA-4 Vax has been evaluated alone and in combination with radiation therapy (RT) and other DNA vaccines (such as PD-1 Vax). The combination of CTLA-4 Vax and RT has shown the ability to enhance effector T cell infiltration into the tumor microenvironment and induce systemic anti-tumor immunity. The vaccine is typically delivered via electroporation to enhance cellular uptake and immunogenicity.
