Overview

CTP-miRNA106a is an investigational peptide-drug conjugate and RNA therapy being developed by Vivasc Therapeutics in collaboration with Georgetown University for the treatment of heart failure and cardiac hypertrophy. The drug consists of a Cardiac Targeting Peptide (CTP) reversibly linked via a disulfide bond to microRNA 106a (miRNA106a), designed to specifically deliver the miRNA cargo to cardiomyocytes. Once delivered to heart cells, miRNA106a downregulates the expression of key genes that contribute to cardiac hypertrophy and inflammation, including Calcium/calmodulin-dependent protein kinase type 2 delta (CaMKIIδ), Phospholipase C beta 1 (PLCβ1), and Histone deacetylase 4 (HDAC4). The drug is administered intravenously and has demonstrated the ability to reverse heart failure parameters in preclinical mouse models. It is currently undergoing in vivo optimization and IND-enabling studies supported by NIH Phase I STTR grants.
