CTP37-DT is a therapeutic cancer vaccine designed to elicit an immune response against human chorionic gonadotropin (hCG), a hormone frequently overexpressed in various epithelial malignancies, including colorectal, pancreatic, and lung cancers. The vaccine consists of a 37-amino acid synthetic peptide representing the carboxy-terminal portion (CTP) of the beta subunit of hCG conjugated to a diphtheria toxoid (DT) carrier. By coupling the self-antigen peptide to the highly immunogenic DT carrier, the vaccine is able to break immune tolerance and induce high-titer anti-hCG antibodies. These antibodies neutralize circulating hCG, which may otherwise act as a growth factor or facilitate immune evasion by the tumor. Clinical studies in patients with metastatic colorectal cancer have demonstrated that the induction of anti-hCG antibodies is significantly associated with improved overall survival.
