Overview

Ctrl-LNP is an experimental control lipid nanoparticle (LNP) used in preclinical research to evaluate the efficacy and specificity of targeted or therapeutic LNP formulations. In the context of atherosclerosis research, it serves as a non-targeted vehicle control to be compared against modified nanoparticles, such as itaconate-conjugated lipid nanoparticles (ITA-LNPs). Studies conducted at Case Western Reserve University have utilized Ctrl-LNP to demonstrate that therapeutic effects—such as the reduction of atherosclerotic plaque size, stabilization of the cholesterol transporter ABCA1, and induction of cholesterol efflux—are specific to the therapeutic payload or targeting moiety rather than the nanoparticle vehicle itself. Ctrl-LNP typically lacks inherent therapeutic activity and does not demonstrate significant accumulation in atherosclerotic plaques.
