Overview

CTS3497 is a **small molecule inhibitor of protein arginine methyltransferase 5 (PRMT5)** being developed for the treatment of **MTAP-deficient advanced solid tumors and lymphomas**. The drug is designed to selectively target tumor cells lacking methylthioadenosine phosphorylase (MTAP), a genetic alteration present in various cancer types. By inhibiting PRMT5, CTS3497 interferes with the methylation of arginine residues on histones and other proteins, impacting gene expression and tumor cell survival. Clinical trials have reported robust and durable anti-tumor responses in MTAP-deficient tumor models. CTS3497 is administered orally and is currently in Phase I/II clinical development, with ongoing studies evaluating safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary anti-tumor efficacy in patients with glioma, non-small cell lung cancer, biliary tract cancer, pancreatic adenocarcinoma, esophageal squamous cell carcinoma, malignant pleural mesothelioma, urothelial cancer, and lymphoma[1][2][3][5][7][9].
