Drug intelligence / Profile preview
CTW0404 is an experimental small molecule that acts as a selective positive allosteric modulator (PAM) of the serotonin 5-HT2A receptor (5-HT2AR). Developed by researchers at the University of Texas Medical Branch (UTMB), CTW0404 is designed to enhance 5-HT2AR signaling without exhibiting intrinsic agonist activity, offering a potential therapeutic strategy for neuropsychiatric disorders and cortical dysfunction. In preclinical studies, CTW0404 demonstrated favorable drug-like properties, including a long half-life, limited cytochrome P450 enzyme inhibition, and high blood-brain barrier penetration with a high brain-to-plasma ratio. Unlike orthosteric 5-HT2AR agonists, CTW0404 does not induce a head-twitch response in animal models, suggesting it lacks hallucinogenic potential.
