CTX-18 is a preclinical-stage molecular glue degrader developed by Captor Therapeutics. It is designed to induce the targeted degradation of G1 to S phase transition 1 (GSPT1), a translation termination factor that is essential for the survival of various cancer cells. By acting as a molecular glue, CTX-18 facilitates the interaction between GSPT1 and an E3 ubiquitin ligase complex, leading to the ubiquitination and subsequent proteasomal degradation of GSPT1. The drug is primarily being investigated for the treatment of high-risk, MYCN-amplified neuroblastoma, as well as other solid tumors including hepatocellular carcinoma, lung cancer, and neuroendocrine (NET) tumors. Preclinical studies have demonstrated significant anti-tumor activity and deep degradation of the target protein in relevant models.
