Overview

CTX-8371 is a fully-human, tetravalent bispecific antibody developed by Compass Therapeutics as a next-generation immune checkpoint inhibitor for cancer immunotherapy. It simultaneously targets and binds to both programmed cell death protein 1 (PD-1) on leukocytes and programmed death-ligand 1 (PD-L1) on tumor cells. Its mechanism of action involves dual blockade of the PD-1/PD-L1 pathway and induction of proteolytic cleavage leading to loss of cell surface PD-1, which may prevent or overcome T cell exhaustion within tumors. Preclinical studies have shown enhanced anti-tumor activity compared to anti-PD-1 or anti-PD-L1 therapies, with additional benefit when combined with agonistic anti-CD137 antibodies. Clinical trials are underway for multiple solid tumors.
