Overview

CTX-B is a potent and selective melanocortin-4 receptor (MC4R) agonist currently under development by **Courage Therapeutics** (also known as nCourage). It is the company's lead candidate designed to treat **MC4R haploinsufficiency (MC4R HI)**, a genetic form of obesity caused by the loss of function in one of the two copies of the MC4R gene. This condition is characterized by early-onset hyperphagia (extreme hunger) and severe obesity, affecting an estimated 100,000 to 150,000 patients in the United States. By acting as a targeted agonist, CTX-B aims to restore MC4R signaling pathways that are critical for regulating energy balance and appetite control. The program is currently in the preclinical stage, addressing a significant unmet need as there are currently no approved targeted therapies specifically for MC4R haploinsufficiency.
