**CTX-MMAE** is an investigational, site-selective epidermal growth factor receptor-targeted antibody-drug conjugate composed of cetuximab conjugated to the antimitotic payload monomethyl auristatin E. It uses a dibromopyridazinedione-based disulfide re-bridging linker system, with an average drug-to-antibody ratio of approximately 3.9. Cetuximab directs the conjugate to epidermal growth factor receptor-expressing tumor cells, while intracellular release of monomethyl auristatin E inhibits tubulin polymerization and causes mitotic arrest. In preclinical KRAS-mutant pancreatic cancer xenograft models, CTX-MMAE showed sub-nanomolar, epidermal growth factor receptor-dependent cytotoxicity, tumor-growth inhibition, and survival benefit without overt toxicity.
