CTX114 is a novel, engineered protein drug developed by Character Biosciences as a complement inhibitor for the treatment of geographic atrophy (GA), an advanced form of dry age-related macular degeneration (AMD)[1][3][4][7]. CTX114 is based on the endogenous complement regulator FHL-1, with enhanced SCR7-mediated ligand binding and improved regulatory potency compared to native CFH or FHL-1[3]. Its mechanism involves potent inhibition of complement activation on retinal surfaces, thereby slowing retinal cell death and vision loss associated with GA in AMD[3][8]. The drug was designed using genetic insights linking variants in Complement Factor H (CFH) and FHL-1 to increased risk of AMD. CTX114 demonstrates >200-fold improved binding to heparin and MDA relative to CFH/FHL-1, as well as superior C3b binding affinity[3]. It is currently being evaluated in early clinical development for its ability to slow progression of GA while preserving vision[2][7].
