CubiCAR is a proprietary, multi-functional chimeric antigen receptor (CAR) architecture developed by Cellectis to optimize the safety and production of allogeneic CAR-T cell therapies. This "all-in-one" molecular design incorporates a specialized extracellular domain that functions as a purification tag, a detection marker, and a safety "off-switch." Specifically, the architecture embeds a CD20-based epitope that allows for the selective depletion of CAR-T cells in vivo through the administration of the anti-CD20 monoclonal antibody rituximab, providing a mechanism to manage potential toxicities such as cytokine release syndrome or off-target effects. Additionally, the tag facilitates the streamlined manufacturing of high-purity CAR-T cell products by allowing for the positive selection of CAR-expressing cells. CubiCAR is a foundational component of Cellectis's UCART (Universal CAR-T) pipeline, integrated into candidates targeting antigens such as CD123, CD22, and SLAMF7 for various hematological malignancies.
