CUE-300 refers to a series of engineered recombinant fusion protein biologics developed by Cue Biopharma for the treatment of autoimmune diseases. These molecules are designed to selectively inhibit autoreactive CD4+ T cells by delivering inhibitory signals and/or enhance the activation and expansion of antigen-specific regulatory T cells (Tregs), thereby controlling aberrant immune responses. The platform leverages Cue Biopharma’s Immuno-STAT technology to present disease-relevant antigens (such as proinsulin in type 1 diabetes) alongside immune-inhibitory ligands like PD-L1. For example, CUE-301 is tailored to target proinsulin-reactive CD4+ T cells in type 1 diabetes using a class II HLA molecule loaded with a proinsulin peptide and fused with two copies of PD-L1 to attenuate pathogenic T cell activity[2][3][4]. The overall goal is precise modulation of antigen-specific overreactive T cells without broadly suppressing the immune system.
