CUE-301 is a preclinical-stage recombinant fusion protein developed by Cue Biopharma for the treatment of Type 1 diabetes. It is part of the company's Immuno-STAT (Selective Targeting and Alteration of T cells) platform, which is designed to selectively modulate the immune system by targeting specific T cell populations. CUE-301 is engineered to address the autoimmune destruction of pancreatic beta cells by targeting proinsulin-specific autoreactive T cells. The molecule's mechanism of action involves the presentation of a proinsulin-derived peptide within an MHC complex to engage the T cell receptor (TCR), while simultaneously delivering inhibitory or apoptotic signals through PD-L1 (targeting PD-1) and a DR4 ligand (targeting Death Receptor 4). This dual-signal approach is intended to induce immune tolerance, exhaustion, or deletion of the pathogenic T cells, potentially halting the progression of Type 1 diabetes without broad systemic immunosuppression.
