CUG172 is a long-acting triple agonist peptide targeting the glucagon-like peptide-1 receptor (GLP-1R), gastric inhibitory polypeptide receptor (GIPR), and glucagon receptor (GCGR). Developed by CureGen, the agent is designed to leverage the synergistic effects of these three metabolic pathways to achieve superior weight loss and metabolic improvement compared to mono- or dual-agonists. By activating GLP-1 and GIP receptors, CUG172 enhances insulin secretion and suppresses appetite, while glucagon receptor activation increases energy expenditure and promotes lipid metabolism in the liver. It is primarily being investigated for the treatment of obesity and metabolic dysfunction-associated steatohepatitis (MASH).
