**CuGem-TSL** is an investigational, thermally activatable liposomal formulation of the nucleoside analog chemotherapy **gemcitabine**. Gemcitabine is complexed with copper(II) gluconate inside temperature-sensitive liposomes, enabling substantially higher drug loading and improved intraliposomal stability relative to non-copper gemcitabine liposomes. Following intravenous administration, localized ultrasound-mediated hyperthermia is intended to trigger rapid gemcitabine release in heated tumor vasculature, increasing intratumoral drug exposure while limiting systemic toxicity. It has shown antitumor activity in murine breast cancer and pancreatic ductal adenocarcinoma models.
