cupabimod is a synthetic oligonucleotide transcription factor decoy that binds and sequesters the nuclear factor kappa-B (NF-κB) complex, thereby preventing NF-κB from interacting with its consensus DNA sites and downregulating expression of multiple pro‑inflammatory and catabolic genes. It was initially developed by AnGes (also referred to as AnGes MG), with Shionogi as a development partner, and has advanced to phase 3 trials in atopic dermatitis and early‑stage clinical development in intervertebral disc degeneration and chronic discogenic low back pain, where single intradiscal injection of the formulation AMG0103 has shown sustained reductions in pain and improvements in disc height. As a transcription factor decoy oligonucleotide, cupabimod represents a targeted gene‑regulatory approach aimed at modulating NF‑κB‑driven pathology in inflammatory and degenerative conditions.
