Cur-2FA is a targeted curcumin-folate conjugate designed to overcome the poor bioavailability and lack of specificity of native curcumin. It consists of one curcumin molecule conjugated to two folic acid molecules, making the compound hydrophilic and enabling targeted delivery to cancer cells that overexpress folate receptors. In preclinical studies, Cur-2FA demonstrated the ability to selectively downregulate HPV 18 transcription and Activator Protein 1 (AP-1) binding activity, specifically reducing the expression of c-fos and fra-1. It has shown potent cytotoxic effects and induction of apoptosis in cervical and breast cancer cell lines, with enhanced bioavailability and a longer half-life in mice compared to native curcumin.
