CUR-N399 is a first-in-class, orally administered small molecule inhibitor of phosphatidylinositol 4-kinase beta (PI4KB), developed by Curovir. PI4KB is a host cell enzyme located in the Golgi apparatus and is essential for the replication of enteroviruses (EVs) and other related viruses. By inhibiting PI4KB, CUR-N399 disrupts viral replication across a broad spectrum of EV subgroups (EV-A, EV-B, EV-C, EV-D), rhinoviruses (RV-A and RV-B), and has demonstrated potent antiviral effects in vitro against respiratory syncytial virus (RSV). Preclinical studies show that it reduces viral load and virus-induced inflammation in multiple infection models. The drug has completed Phase 1 clinical trials evaluating its safety, tolerability, and pharmacokinetics in healthy adults. It is being developed primarily for the treatment of enterovirus infections but also shows potential against RSV and other respiratory viruses[2][3][10].
