Drug intelligence / Profile preview
LipoCurc (liposomal curcumin) is an investigational intravenous formulation of curcumin encapsulated in lipid nanoparticles, developed by SignPath Pharma to overcome the poor bioavailability and rapid metabolism of standard curcumin. The drug acts as a multi-target immunomodulator and anti-tumor agent. Its mechanism of action involves the inhibition of multiple immunosuppressive pathways, including the blocking of PD-L1, PD-L2, and CTLA-4 signaling, as well as the reduction of indoleamine 2,3-dioxygenase (IDO) production. Furthermore, LipoCurc inhibits the overexpression of hypoxia-inducible factor 1 (HIF-1), which is often driven by IDH1 mutations in glioblastoma cells. It is currently being evaluated in Phase I/II clinical trials (such as NCT05768919) in combination with radiotherapy and temozolomide for the treatment of newly diagnosed high-grade gliomas, including glioblastoma multiforme (GBM). The FDA has granted LipoCurc Orphan Drug Designation for the treatment of malignant glioma.
