Overview

Curcumin-conjugated magnetic nanoparticles (Cur-MNPs) are engineered nanoparticles composed of superparamagnetic iron oxide (SPIO) magnetic nanoparticles conjugated with curcumin. These nanoparticles are coated with polyethylene glycol-polylactic acid block copolymer and polyvinylpyrrolidone, resulting in stable and biocompatible particles with a mean diameter of less than 100 nm. They are designed for the non-invasive diagnosis of Alzheimer's disease (AD) by detecting amyloid plaques using Magnetic Resonance Imaging (MRI). Curcumin specifically binds to amyloid plaques, and the magnetic properties of the SPIO allow for visualization via MRI, offering a non-radioactive alternative to positron emission tomography (PET). Cur-MNPs have demonstrated no cytotoxicity in Madin-Darby canine kidney (MDCK) or differentiated human neuroblastoma cells (SH-SY5Y) and have shown the ability to cross the blood-brain barrier in an in vitro model. They have been shown to co-localize with amyloid plaques in ex vivo T2*-weighted MRI of Tg2576 mouse brains after injection.
