Cusatuzumab is a humanized IgG1 monoclonal antibody that targets CD70, a ligand expressed on activated lymphocytes and mature dendritic cells. By binding to CD70, cusatuzumab blocks the CD70-CD27 signaling pathway involved in malignant cell proliferation and survival. This blockade inhibits leukemic stem cell proliferation, reduces leukemic blast cells, impairs regulatory T-cell survival (preventing tumor immune escape), and restores normal myeloid differentiation. Cusatuzumab also induces direct Fc-mediated effector functions such as enhanced antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis. It is under investigation primarily for hematological malignancies including acute myeloid leukemia (AML) and high-risk myelodysplastic syndrome (MDS), especially in patients unsuitable for intensive chemotherapy[1][3][5][8].
