Overview

**Cusatuzumab + azacitidine** is a combination of a monoclonal antibody and a hypomethylating agent, investigated primarily for the treatment of newly diagnosed acute myeloid leukemia (AML) in patients ineligible for intensive chemotherapy. Cusatuzumab is a high-affinity humanized IgG1 monoclonal antibody targeting CD70, a protein overexpressed on AML blasts, leading to anti-leukemic effects via enhanced antibody-dependent cellular cytotoxicity (ADCC) and inhibition of CD70/CD27 signaling. Azacitidine is a small molecule DNA hypomethylating agent that induces cell differentiation and apoptosis by incorporating into DNA and inhibiting DNA methyltransferases. The combination aims to exploit complementary pathways to improve clinical outcomes in AML. Primary clinical studies have shown that the combination is generally well tolerated and demonstrates preliminary efficacy, but further development in some settings has been discontinued due to changes in AML treatment standards.
