**cusatuzumab + venetoclax + azacitidine** is an investigational combination therapy comprising three agents: cusatuzumab, a monoclonal antibody targeting CD70; venetoclax, a small molecule inhibitor of BCL-2; and azacitidine, a hypomethylating agent classified as a DNA methyltransferase inhibitor. This regimen is under development for the treatment of acute myeloid leukemia (AML), particularly in patients who are unfit for intensive chemotherapy. Cusatuzumab is designed to block the CD70 pathway and induce direct cytotoxicity in leukemic stem cells, venetoclax promotes apoptosis by inhibiting the anti-apoptotic protein BCL-2, and azacitidine causes DNA hypomethylation, leading to reactivation of silenced genes that suppress leukemia cell growth. This triple combination is being studied clinically to explore potential synergistic activity and to improve remission rates and survival outcomes over standard dual therapies.
