Custirsen is a second-generation antisense oligonucleotide designed to inhibit the production of clusterin, a cytoprotective chaperone protein that is overexpressed in many cancers and associated with treatment resistance. By binding to the mRNA of the clusterin gene, custirsen reduces clusterin protein levels, thereby sensitizing tumor cells to chemotherapy and promoting apoptosis. It has been primarily investigated as an adjunct therapy for metastatic castration-resistant prostate cancer and advanced or metastatic non-small cell lung cancer. Custirsen is administered intravenously and has been studied in combination with chemotherapeutic agents such as docetaxel or cabazitaxel. The drug remains investigational and has not received regulatory approval.
