CV301 + capecitabine + durvalumab is an investigational combination therapy for cancer. It consists of: - **CV301**: A poxviral-based therapeutic cancer vaccine using recombinant Modified Vaccinia Ankara (MVA) and Fowlpox (FPV) viruses encoding tumor-associated antigens carcinoembryonic antigen (CEA), mucin 1 (MUC1), and a triad of co-stimulatory molecules known as TRICOM (ICAM-1, LFA-3, B7-1). The vaccine is designed to prime and boost the immune system to recognize and attack tumor cells expressing these antigens[1][2][3]. - **Capecitabine**: An oral prodrug that is converted to 5-fluorouracil in the body; it inhibits DNA synthesis by interfering with thymidylate synthase activity. - **Durvalumab**: A human monoclonal antibody targeting programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 on T cells. This releases inhibition of T-cell mediated anti-tumor immunity[6][8][10]. The combination aims to enhance anti-tumor immune responses through both direct cytotoxic effects from chemotherapy and immune checkpoint blockade, while the vaccine component primes specific T-cell responses against tumor antigens.
