Overview

The RNActive rabies vaccine refers to a series of messenger RNA (mRNA)-based prophylactic vaccine candidates developed by CureVac AG, specifically **CV7201** and **CV7202**. These vaccines utilize CureVac's proprietary RNActive platform, which employs unmodified mRNA encoding the rabies virus glycoprotein (RABV-G). CV7201, the first-in-human formulation, utilized a cationic protein (protamine) for mRNA complexation, while the subsequent CV7202 utilized lipid nanoparticles (LNPs) for encapsulation. The vaccine works by delivering mRNA into host cells, which then produce the rabies virus glycoprotein, thereby inducing a protective immune response and the generation of neutralizing antibodies. While both candidates reached Phase 1 clinical trials, they have largely been superseded by CureVac's second-generation and chemically modified mRNA technologies developed in partnership with GSK.
