Overview

CV8102 is a synthetic, noncoding, uncapped single-stranded RNA (ssRNA) containing several polyU-repeats and complexed with a polymeric carrier formed by disulfide-crosslinked cationic peptides. It acts as an immunostimulatory agent by activating innate immune receptors such as toll-like receptor 7 and 8 (TLR7/8) and retinoic acid-inducible gene I (RIG-I), mimicking viral infection in the tumor microenvironment. This activation leads to the recruitment and activation of antigen-presenting cells at the site of injection, promoting presentation of tumor antigens to T cells in draining lymph nodes. The result is an enhanced anti-tumor immune response both locally at injected tumors and systemically against distant lesions or metastases. CV8102 has been investigated as monotherapy or in combination with PD-1 inhibitors for various cancers including cutaneous melanoma, cutaneous squamous cell carcinoma, head and neck squamous cell carcinoma, adenoid cystic carcinoma, and hepatocellular carcinoma.
