Overview

CVAX-01 is an intranasal COVID-19 vaccine candidate developed by CastleVax, a clinical-stage spin-off from the Icahn School of Medicine at Mount Sinai. The vaccine utilizes a live-attenuated Newcastle Disease Virus (NDV) vector platform to express a stabilized version of the SARS-CoV-2 spike protein. Unlike traditional intramuscular vaccines, CVAX-01 is administered via a nasal spray to induce mucosal immunity (secretory IgA) in the upper respiratory tract, aiming to block viral entry and reduce transmission in addition to preventing severe disease. In 2024, the program received significant backing, including a $34 million grant from BARDA to fund a 10,000-participant Phase 2b trial, and support from the New York State Biodefense Commercialization Fund. The vaccine is delivered using AptarGroup's specialized intranasal delivery devices.
