Overview

CVD 1902 is a live attenuated oral vaccine candidate against Salmonella enterica serovar Paratyphi A (S. Paratyphi A), which causes paratyphoid fever. It was constructed from wild-type parent strain S. Paratyphi A strain ATCC 9150 by deleting the guaBA chromosomal operon (involved in guanine nucleotide biosynthesis) and the clpX gene (encoding a chaperone ATPase). The clpX mutation results in hyperexpression of flagella and serves as a second attenuating mutation to minimize the risk of reversion to full virulence. The vaccine is designed to stimulate both local protective immune responses along the intestinal surface and systemic immunity.
