CVGBM is an investigational therapeutic mRNA-based cancer vaccine developed by CureVac (acquired by BioNTech) for the treatment of glioblastoma (GBM). The vaccine consists of nucleotide-unmodified mRNA encapsulated in lipid nanoparticles (LNPs). It encodes a fusion protein containing eight epitopes derived from tumor-associated antigens (TAAs) that are over-presented in GBM, including five restricted to HLA-A*02:01 and three restricted to various HLA-DR alleles. CVGBM is designed to induce both CD8+ and CD4+ T-cell responses against the tumor. It is currently being evaluated in Phase 1 clinical trials (NCT05938387) for patients with newly diagnosed, surgically resected, MGMT-unmethylated glioblastoma or astrocytoma with a molecular signature of unmethylated GBM.
