CVL006 is a novel bispecific antibody developed by Convalife for the treatment of advanced solid tumors. It is designed to simultaneously target and inhibit both programmed death-ligand 1 (PD-L1) and vascular endothelial growth factor A (VEGF-A). By fusing an anti-PD-L1 VHH domain with a humanized IgG1 anti-VEGF monoclonal antibody, CVL006 blocks the PD-L1/PD-1 immune checkpoint pathway and VEGF-driven angiogenesis. This dual mechanism aims to suppress tumor-induced immunosuppression while also inhibiting tumor vascularization, thereby enhancing antitumor efficacy. Preclinical studies have shown that CVL006 has superior antitumor activity compared to monotherapy with either PD-L1 or VEGF inhibitors alone and outperforms approved bispecific antibodies such as AK112 (ivonescimab) in animal models. The drug retains antibody-dependent cellular cytotoxicity (ADCC) functionality and is currently being evaluated in Phase I clinical trials for safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced solid tumors[4][7][1].
